Cannot Penetrate Multiple Layers is an attack behavior characterized by not being able to penetrate multiple layers at all, not even when given damage buffs or when attacking Debuffed Bloons, and only does more than one damage on multi-HP single layers. It is part of the damage types family. In Bloons TD 6, this was a previously lesser known game mechanic that had secretly gotten removed from many attacks as of Version 31.2. It was likely removed in Bloons TD 6 to minimize redundant damage calculations.
